Despite the fact that crystals of this protein were obtained in 1976 and a low-resolution structure determined to 6 A resolution was published in 1984, determination of a high-resolution structure had proved elusive. For this reason heteronuclear, multi-dimensional NMR was carried out to determine the structure of this protein. 15 N- and 15 N13 C-labelled KSI samples were obtained from bacteria grown in media rich in 15 N and 13 C. Ten percent dioxane was included in solvent to inhibit aggregation at concentrations greater than 80 M. Four-dimensional 15 N, 13 C- and 13 C, 13 C NOESY spectra were generated and used to assign backbone and side-chain signals. A list of 1865 distance constraints ((1) below) was used as an input to generate 20 individual distance geometry structures which are shown in (2). Disorder at the dimer interface is responsible for the greater convergence of the family of monomeric structures compared to that for the dimer. The model agreed well with previously published structure function work on this enzyme and allowed identi cation of important active-site residues.
